It is always a pleasure getting the chance to speak with Doug Drysdale, CEO of Cybin Inc, and especially now under the current circumstances. Cybin has recently initiated PARADIGM (the company’s phase 3 program), and has released 12 month data from their phase 2 trial looking at CYB003 as an adjunct treatment in major depressive disorder (MDD). As far as I am aware, Cybin is the first company in the psychedelic drug development space to release 12 month data. The data was incredibly encouraging for both the 12 mg and 16 mg cohorts. In the 16 mg group, 7/7 trial participants who received two doses of CYB003 (dosing separated by three weeks) were responders to treatment (50% or more reduction in depressive symptoms), 12 months later. 5/7 trial participants were in remission (MADRS score ≤10). The other two responders in the 12 month follow up had a MADRS score of 11. To see trial participants with moderate-severe depression experience a mean reduction of 23 points on the MADRS, 12 months after treatment, is remarkable. Topics of this interview/discussion include: • Cybin’s phase 3 program (PARADIGM) • phase 2 CYB003 12 month data • The STAR*D study which showed that treatment response and remission rates get lower and lower, treatment after treatment, for patients suffering from depression. (Too many patients are failed by what is currently available today). • What line of treatment CYB003 might be prescribed for in the future, if approved? • Positive depression data from more traditional drug development companies in the neuropsychiatry space. (Improvements over SSRIs/SNRISs, but where is the remission data?) How should we look at data with psychedelic compounds compared to more traditional treatment approaches? • Psychedelic compounds having the potential to change the course of illness for many patients, going beyond symptom management. • Upcoming phase 2 data readout of CYB004 in generalized anxiety disorder, anticipated in Q1 2025 (CYBOO4 is deuterated N,N-DMT IM formulation. Rapid onset of effects, 2-3 minutes, with total duration of psychedelic effects expected to last around 90 minutes) • RFK Jr.’s comments made about psychedelics. (RFK Jr. and many in the new administration seem to be “pro psychedelics” but will they be pro psychedelic drug development?) Congratulations to Doug and the entire team at Cybin on their incredible execution the past several years. I am really looking forward to following the progress.
