Monoclonal antibody-based therapeutics are well established in the pipelines of biopharmaceutical organizations and have decades of development ahead. However, not all of them have disclosed or confirmed amino acid sequences. Ultra-high-performance liquid chromatography mass spectrometry provides a powerful tool to obtain the sequence information to the residue level. Historically, the process has needed expert users and therefore was extremely low throughput and is often outsourced.

Now, by using the right practical tools, de novo sequencing now can be easily turned into an in-house, higher throughput procedure with extremely high confidence. Advances in sample preparation have allowed for automated digestion strategies using various proteolytic enzymes while removing roadblocks for orthogonal protein sequencing strategies—ultimately speeding result generation. Leveraging different fragmentation techniques on a high-end high-resolution accurate mass spectrometer, followed by easy to use software tools, makes this whole analytical process as simple as possible.

In this GEN webinar, our presenters will demonstrate that almost 100% of the sequence information is correct in the samples analyzed when tested against multiple (known) proteins. Our presenters will also demonstrate how challenging regions can be identified rapidly by users so that they know immediately whether the challenge lies at the sample prep, data paucity, or instrument level.
